Long- Term Results of Extraocular Muscle after Subconjunctival Gentamicin Injection in Rabbit



Abstract
It was established that subconjunctival injection of gentamicin causes an acute toxic myopathy on extraocular muscle in rabbit.
Light microscopic findings of the acute toxic myopathy were focal infilterations of the polymorphonuclear leukocytes. After few days, instead of polymorphonuclear leukocytes, mononuclear leukocytes infilteration were noted with muscle fiber
degeneration.
Electronmicroscopic findings of acute toxic reactions were lysosomal overloading in fibroblast caused by gentamicin.
But no data are available about long-term results of extraocular muscle after subconjunctival gentamicin injection and the reversibility of gentaminin-induced myopathy.
So, we performed enucleation of rabbits at 24 hours, 1 week, and 12 weeks after sunconjunctival gentamicin injection(40mg%)
@ES The follwing results wee obtained.
@EN 1. Light microscopic findings on 1 day and 1 week were those of acute toxic myopathy.
2. Light microscopic findings of muscle on 12 weeks were nearly normal without inflammatory cells or fibrosis.
3. Electronmicroscopic findings of the specimen on 12 weeks were also nearly normal except enlargement of sarcoplasmic reticulum and disruption of mitochodrial cristae. These findings seemed to be the evidences of regeneration of the muscle.
All of these results suggest that gentamicin-induced myopathy are reversible just like gentamicin-induced toxicity of other organs.
